The accident happened on the A64 at Whitwell Hill – the main route between York and Scarborough.
The crash happened on the east-bound carriageway at about 12.30am on Sunday and involved a black Mercedes.
"It is believed the car was in the outside lane of the dual carriageway when it collided with the barrier in the central reservation and the vehicle overturned," said a North Yorkshire Police spokesman.
"The driver, a 26-year-man from the Malton area, was sadly pronounced dead at the scene.
"Three passengers were taken to York District Hospital by ambulance. A 22-year-old woman from Malton with serious leg injuries, an 18-year-old woman from Malton with neck and spinal injuries, and a 29-year-old man from the Knaresborough area with cuts and bruises.
"The section of road was closed until around 7am on Sunday while the emergency services tended to the casualties and to allow the police to carry out a full examination of the fatal collision scene."
